AdaptX MPF Derisking Innovation: Spotlight Asymmetric Dialysis

In designing the AdaptX MPF product line our N+1 engineers wanted to do our part to address this specific CAPEX challenge that our customers face. Ultimately helping our customers derisk their pathways to innovation and next generation operational benefits.
Today we will highlight the AdaptX MPF Asymmetric Dialysis operational mode a next generational alternative to traditional UF/DF unit operations.
AdaptX offers both the known and classical process, recirculated UF/DF, and the next generation Asymmetric Dialysis as both out of the box standard operational modes. Allowing you to chose next generation innovation or classical unit operations with no additional CAPEX commitment following the initial purchase of the AdaptX MPF System.
Pioneered and introduced for next generation mAb processes by Patil, Chen, and Coffman of Astra Zeneca Biologics and in published work from Yehl, Jab, and Zydney in the examination of purification of IgG, Asymmetric Dialysis is a one-step continuous process designed to eliminate the need for a traditional two step batch UF/DF operations.
Careful manipulation of the inlet and outlet flowrates on the retentate and permeate side of the filter, Asymmetric Dialysis has been demonstrated to achieve desired product concentrations and salt removal in a single step using up to 75% less diafiltration buffer than a traditional two step UF/DF batch process.
In this process the dialysis buffer is introduced on the permeate side (shell side) of the hollow fiber filter in a counter current direction to the inlet feed flow direction on the lumen side of the filter and with only the use of modest TMP.
Unlike other market alternatives, AdaptX MPF’s comes standard with the necessary number of low shear pumps on the retentate and permeate system along with the necessary componentry in its four modular zones on the filter side face of the system necessary to perform Asymmetric Dialysis single pass filtration process. Figure 1 displays the connected modular flow kit segments for the Asymmetric Dialysis Unit Operation Configuration on the AdaptX MPF System.

Another benefit of the AdaptX MPF System, over pump only flow control solutions, is the flow control achievable through use of the Equilibar SDO valve on both the permeate side valve on both the permeate side and retentate side of the filter to deliver the flow turn down ratios necessary to finely modulate the flow conditions and TMP during the Asymmetric Dialysis Process within the AdaptX patented modular configuration blocks. This control valve coupled with the low shear pumps from Levitronix on the retentate and permeate sides of the filter allow the AdaptX to offer customers a linearly scalable, low shear system, with fine tune flow control capabilities specifically designed for this unit operation’s optimization.
